Microscopy methods for morpho-functional characterisation of marine invertebrate haemocytes
نویسنده
چکیده
In this chapter, a useful list of well-established morphological and functional protocols is provided, to facilitate the design of experiments in the field of the characterisation of haemocytes of marine invertebrates, which have been emerging in the last decade as model organisms in ecotoxicology, comparative immunology, phylogenesis and evolution. This treatment offers a brief collection of methods and procedures for both light and electron microscopy, most of which were previously developed for vertebrates and therefore require modifications and adjustments. Many suggestions to solve common difficulties and errors are presented and clearly explained, covering haemocyte collection, cell culture, techniques of staining, and enzyme histochemistry and immunohistochemistry to obtain the best results.

Cytochemical properties of Botryllus schlosseri haemocytes: indications for morpho-functional characterisation.
In the present study, we carried out a detailed light microscopy investigation of the cytochemical properties of the haemocytes of the colonial ascidian Botryllus schlosseri, using new cytochemical stains and enzymatic markers, a panel of antibodies and lectins as probes to characterise Botryllus blood cells further. Results indicate that lymphocyte-like cells are circulating undifferentiated c...
متن کاملBenthic invertebrate community in Khur-e-Mussa creeks in northwest of Persian Gulf and the application of the AMBI (AZTI\'s Marine Biotic Index)
Benthic invertebrates are a well-established target in environmental quality status evaluations. The AMBI (AZTI's Marine Biotic Index) was developed to determine the impacts and the quality status of soft-bottom marine benthic communities. The aim of this study was investigating multivariate pattern of benthic invertebrate community and application of AMBI (AZTI's Marine Biotic Index) for...
متن کاملFirst cytochemical study of haemocytes from the crab Carcinus aestuarii (Crustacea, Decapoda)
For the first time, a morphological study of haemocytes from the crab Carcinus aestuarii was carried out by means of light microscopy and differing cytochemical assays. Analysis of haemocyte size frequency distribution (performed by means of a Coulter Counter) revealed the presence of two distinct haemocyte fractions in C. aestuarii haemolymph, depending on cell size. The first fraction was of ...

متن کاملFunctional characterisation of phagocytes in the Pacific oyster Crassostrea gigas
Invertebrates lack canonical adaptive immunity and mainly rely on innate immune system to fight against pathogens. The phagocytes, which could engulf and kill microbial pathogens, are likely to be of great importance and have to undertake significant roles in invertebrate immune defense. In the present study, flow cytometry combined with histological and lectin staining was employed to characte...
